Unfortunatly there is no quick answer to your quick question. A dead cylinder can be caused by any number of things. Some simple some not so simple. In order for your engine to perform it must possess three elements:
1) Spark
2) Compression
3) Fuel
The lack of any one of these will cause malfunction. So divining this mystery requires some mechanical detective work. Since you already suspect spark (the deiseling or running on often indicates timing) I would suggest you replace the Spark Plug wire and Spark Plug, set your timing, and see if there is a change.

BTW: Our little cars seem to insist on frequent valve adjustment. Listen closely to #1 for a tapping sound.
