Say I just washed my car at the coin-operated wash (brushless) and drove it home. What can I do to get the water spots off and really make it shine?

Search eBay for 'california blade' & 'california duster'. I carry both in the trunk and the blades are great after a brushless wash. Takes a few minutes, but is a nice bonding experience with my 7 ;-)
The duster is very good at grabbing whatever floats onto the paint within minutes of compleeing a thorough washing ;-)

I use windex on the windows and chrome. That is my choice for glass as well although there may be better (more expensive) solutions.
Is there a spray to use lightly on the car itself and wipe it down?
Haven't settled on anything for that yet, let's see what the experts say.

What's the best material to buff the car?

Haven't gotten to that one either although a random orbital buffer (best pad material?) is on my list if I find one on sale.
