Robbie,
I believe you are refering to the screw (actually, it's a small 10mm bolt) on the top of the thermostat housing. Fill the resevoir with fluid when the car is cool. Start the car. (You can leave the top off the resevoir while the car is warming up.) Make sure your heater is on also, so the heater core will fill.

Now, loosen this bolt slightly, and you should see coolant gurgle from the slot below. Top off the resevoir if you have to, screw the top on, and as the car continues to warm up, you should eventually see a steady flow of coolant (no air) from the slot. Tighten the screw, and you're done.

Also, make sure you monitor your temp gauge as you do this to make sure everything's ok.
